DEVELOPMENT OF GASTRORETENTIVE SYSTEMS FOR FAMOTIDINE: IN VITRO CHARACTERIZATION

India Floating tablets of famotidine were developed to prolong gastric residence time, leading to an increase in drug bioavailability. Tablets were prepared by direct compression technique, using polymers hydroxypropyl methylcellulose (HPMC K4M and HPMC K15M). Tablets were evaluated for physical characteristics, weight, thickness, hardness and friability; drug content, swelling, floating properties and in vitro drug release characteristics for 12 h. Tablets exhibited sustained and prolonged drug release profiles while floating over the dissolution medium. The release pattern followed was anomalous non-fickian type, indicating that water diffusion and polymer rearrangement played an essential role in drug release. The best formulation was selected based on in vitro characteristics and was subjected for in vivo radiographic studies. These studies revealed that the tablets remained in stomach for more than 4.5 h in fed condition indicated that gastric retention time was increased by floating principle.
