RP-UPLC Method Development and Validation for Simultaneous Estimation of Mometasone Furoate and Miconazole Nitrate in Semisolid Dosage Form

DOI : 10.23893/1307-2080.APS.05819 Viewed : 22218 - Downloaded : 3827 An innovative, rapid and precise RP-UPLC method was developed and validated as per ICH guidelines for simultaneous estimation of Mometasone furoate (MF), and Miconazole nitrate (MN) in topical dosage form. Chromatographic separation was carried out using Agilent C18(4.6mm×100mm, 5µm) column and mobile phase consists of 0.1% v/v triethylamine: methanol: acetonitrile (40:30:30 V/V/V; pH 3.5). The flow rate was 0.6mL/min and detection was set at 235 nm in UV detector. Retention time of MF and MN were 0.59 min and 1.13 min respectively. The method shows good linearity over the concentration range of 10-30 µg/mL MF and 200-600µg/ ml MN. Recovery for both analytes was found to be 99.58% and 98.51% respectively. LOD and LOQ for MF and MN were found as 5.452 and 0.501µg/ml, 1.485 and 1.20µg/ml respectively. This newly developed RP-UPLC method can be successfully applied for simultaneous determination of MF and MN in topical dosage form.
